Each of the eight characters are located somewhere within the levels of the single player game. If you find a character, blow them away. After you successfully kill them, you will be able to select them on the Biker select screen. Another way to do this is to unlock Battle Mode and hope that a CPU opponent shows up as one of the eight secret characters so that you don't have to search for them in the single player game.
