In the first level, a yellow icon with a hand appears above a clock/mirror - what does it mean and how do I get over the fences to the rest of the level.

It's the mirror that nararates the story and the hand means to press the action button whatever you've configued it to be. The open the gate I think you have to become Donkey and buck the gate open or find a lever or something.
