It'll be awhile before you get the sixth locator card. Forget Mai, for she won't put up her locator card. Defeat Arkana at the card shop (not Grampa's),then head to Grampa's shop to talk to Namu (Malik), and the Bridge and Building will be available to you. Head to the Building (that's where Mai is) to level up some more if you want as well as beating Joey. Head to the Bridge and head right (left for your character) after talking to Kaiba and defeat Strings. Afterwards, the Aquarium should be available. Defeat all the duelists around the aquarium before taking on Mako, because he won't duel you unless you do, including Joey. Once you're done, challenge him and defeat him for the sixth locator card.
