I encountered my first two Little Sisters the other day. The first one was by herself and I decided to save her.
For the second I had to kill a Big Daddy, and afterwards I figured I did all that work to kill him that I would 'kill' the Little Sister to get more ADAM.
My question is, since I've killed a Little Sister does saving them now get me any rewards from that one lady? (Can't remember her name) Or does the fact that I've kill one make it so she won't reward me at all?

I'd like to correct some numbers here. You do not get more ADAM for Rescuing all the little sisters. Take a look at the numbers:
Rescue 3 Little Sisters x 80 ADAM for each = 240 ADAM Total 240 ADAM + 200 ADAM from Tenenbaum's Gift = 440 ADAM Total 440 ADAM x 7 groups of 3 Little Sisters = 3080 ADAM Total
Final result: Rescuing all Little Sisters gives you 3080 ADAM.
Harvest 3 Little Sisters x 160 Adam for each = 480 ADAM Total No extra Adam from Tenenbaum 480 ADAM x 7 groups of 3 Little Sisters = 3360 ADAM
Final result: Harvesting all Little Sisters gives you 3360 ADAM.
Now, unless you're really, really bad at math, 3080 =/= 3360. You actually get an extra 280 ADAM for Harvesting all the Little Sisters. However, you'll miss out on the 'Hypnotize Big Daddy' Plasmids, as well as the Armored Shell 2 Plasmid, as those can only be gotten from Tenenbaum's Gifts.
Frankly, if you know what you're going for, and use your ADAM wisely, the ADAM you get from Rescuing is more than enough to pick up what you need. You'll have to do some picking and choosing early on, but if you're patient, you can survive.
And for the record, I Rescue them. To Harvest them goes against my nature. Just because they've been taught to do horrible things, that doesn't mean they're no longer people. If they can be rescued, then they should be. And the video you get at the end proves that it's not futile. So yes, I do it for the warm fuzzies.

im gonna add a bit of a spoiler to mine here. Not only do you get more adam but you get a hypnotize big daddy plasmid. Title says it all.You can trick a big daddy into temporarily thinking your a little sister. =D

I harvested all of them in my normal mode playthrough,but I'll rescue all of them on hard mode.

It's been a long time since I've last posted here. (Performs level ten scanning sweep) A long time. Yet... only a few new people.
Still, I've recently picked up Bioshock after reading my Game Informer article on Bioshock 2: Sea of Dreams. I have to say, I nearly had an overload from it. In Bioshock, you don the armored diving suit of the Big Daddy, Bioshock 2? Well, I won't drop any spoilers here, but I will say I've always wanted that drill or the rivet gun. I'm going to damage a few of my manipulator motivators (The bones in fingers to you biologicals) when I finally get my manipulators (Hands) on my copy.
Anyway, back on topic, I've recently picked up my copy of Bioshock and I've still been unable to harvest even a single Little Sister. It breaks my powercore to harm the innocent for any reason, and in some other games I have found myself sacrificing them for various ends if that ends was justified. I'm just so happy when I save them, but I feel hollow at the end, when I think of what is to come. You should check out the Big Daddy stories on Fanfiction.net, they're quite 'Riveting' (Sorry for the pun. I could not stop myself.) when you get into it. You'll look at Rapture in a whole new light, I guaran-frickin'-tee it.
and you get that fuzzy, warm feeling inside when you look into the eyes of the rescued little sister lol

Well if you like being evil, harvest the little sisters, and if you are nice, save them. However, I read somewhere that you gain more ADAM in the game when you harvest them.

I've read that if you harvest more than one, Jack (The guy you play as) takes over rapture and uses all of the Adam in a manner similar to Fontaine.
Also, I can't bring myself to harm the innocent so I save the Little Sisters. The Big Daddys however are my paychecks. I see a Big Daddy after I've saved all of the Little Sisters in an area, I'm going to kill them and take their money and/or invent parts. They are neutral not innocent so by my logic they are officially target practice.
