The most lethal, yes; but I don't consider it the best due to the fact of it drawing the fun and skill out of the game.
I like the Iron shield combo, or the mighty .. overlooked .. overpowering .. alucart shield combo!

The best shield rod combo is the crissaegrim/alucard shield glitch.
Equip a sheild rod and Alucard shield, activate the special attack and press pause. Switch the shield rod with a crissaegrim and un-pause. The shield will still do 199 dmg. 5 times per second and you can tap both buttons very quickly, to get insane amounts of damage.
